A barcoded variant of TnSeq (RB-TnSeq) was used to perform hundreds of in vitro genome-wide fitness assays in B. thetaiotaomicron. The researchers further profiled fitness during mono-colonization of germ-free mice. The rich dataset generated links genes that previously lacked informative annotations to specific processes including polysaccharide degradation, disaccharide catabolism, and bile salt tolerance.
